After tragedy strikes the kingdom of Dor; when Roscuro (Dustin Hoffman) the rat falls in the soup and the shocked queen faints and drowns in her bowl of soup, the king banishes rats and outlaws the cooking and eating of soup, beginning the Dark Ages for Dor. Then one day in those dark days Despereaux (Matthew Broderick) the mouse is born, and unlike the other mice he is brave. Yet the mouse council banishes him too when he speaks to a human- the lonely Princess Pea (Emma Watson)- and becomes her friend.

In the dungeon, where rats and mice are banished and ruled by the evil Botticelli (Ciarán Hinds), Despereaux is almost eaten by the rats if it wasn't for Roscuro who saves him. Despereaux tells Roscuro of the sadness that surrounds the princess, and together both of them might just have a shot at saving the princess and maybe even bring back soup to the kingdom of Dor...
